01 / PRODUCT DOSSIER

What 4-Aminophenol is, and why buyers source it

A working overview of the compound's chemistry, its central role in paracetamol manufacture, and the grades Chelora Chemicals keeps in regular supply.

4-Aminophenol is a bifunctional aromatic compound carrying both an amine and a hydroxyl group on opposite ends of the benzene ring, a combination that makes it exceptionally reactive at both positions and, more specifically, makes it the direct chemical precursor of paracetamol (acetaminophen): a single acetylation of the amine group converts 4-aminophenol into the finished analgesic active pharmaceutical ingredient in one step. That one reaction is responsible for the overwhelming majority of global 4-aminophenol demand, making the compound's quality and purity profile a matter of direct pharmaceutical consequence rather than just an industrial convenience.

At room temperature it is a white to light tan crystalline powder that darkens slowly on prolonged exposure to air and light, owing to oxidation at the easily oxidised amine/hydroxyl pair — a sensitivity that governs how the material is packaged and stored far more than any acute hazard does. It is only slightly soluble in cold water but dissolves more readily in hot water and in common polar organic solvents.

Where it fits in the value chain

Pharmaceutical-intermediate-grade 4-aminophenol is acetylated with acetic anhydride to produce paracetamol API, the single largest global use of the compound by a wide margin. A smaller but still significant volume is consumed in photographic developer chemistry, where its combined amine/hydroxyl reactivity suits it to developing-agent synthesis, and in the manufacture of azo and sulphur dyes and rubber antioxidants, where the same bifunctional reactivity is put to different downstream use.

Grades we supply

Chelora Chemicals offers 4-Aminophenol in pharmaceutical-intermediate grade, meeting the assay, heavy-metal and related-substance specifications needed for paracetamol synthesis, and in technical grade for dye and rubber-chemical applications. Each grade is supplied with a batch-wise certificate of analysis.

02 / MANUFACTURING ROUTE

How 4-Aminophenol is produced

An overview of the production route for orientation purposes. Our manufacturing partners' own process parameters and in-process controls are proprietary and are not disclosed beyond what is needed to interpret the certificate of analysis.

4-Aminophenol is produced predominantly by catalytic hydrogenation of nitrobenzene under carefully controlled conditions: rather than reducing all the way through to aniline, the reaction is stopped at the partially reduced phenylhydroxylamine intermediate, which then undergoes an acid-catalysed rearrangement — the Bamberger rearrangement — that migrates the nitrogen-oxygen functionality to give 4-aminophenol directly. Controlling the hydrogenation to stop cleanly at the phenylhydroxylamine stage, without over-reducing to aniline or under-reducing to leave nitroso by-products, is the most demanding step in the whole route and the one that most determines yield and purity.

An alternative, older route reduces 4-nitrophenol directly to 4-aminophenol using iron and acid or catalytic hydrogenation, though the nitrobenzene/Bamberger route is now the dominant commercial process. Either way, the crude product is purified by recrystallisation to reach the assay and colour specification required of pharmaceutical-intermediate grade, since even small amounts of coloured oxidation by-products are unacceptable in material destined for paracetamol synthesis.

03 / APPLICATIONS

Industries and processes that use it

4-Aminophenol is consumed overwhelmingly by paracetamol manufacture, with smaller volumes into photographic and dye/rubber chemistry. The fields below account for most of the demand we see through our export desk.

Field 01

Paracetamol (Acetaminophen) API

Acetylated with acetic anhydride in a single step to produce paracetamol, the world's most widely used over-the-counter analgesic and antipyretic active ingredient.

Field 02

Photographic Developing Agents

Used in the synthesis of photographic developer chemistry, where its combined amine/hydroxyl reactivity is well suited to developing-agent structures.

Field 03

Azo & Sulphur Dye Intermediates

A building block for azo and sulphur dye synthesis, exploiting the amine group's diazotisation reactivity and the phenol group's coupling reactivity.

Field 04

Rubber Antioxidants

Used in the manufacture of certain rubber antioxidant and antiozonant chemicals, where the phenolic/amine structure provides radical-scavenging activity.

04 / REACTION CHEMISTRY

How the molecule is typically carried forward

The bifunctional amine/hydroxyl reactivity governs almost all of 4-Aminophenol's further reaction chemistry. The categories below account for most of the ways it is converted into a finished product.

Acetylation (to Paracetamol)

The amine group is selectively acetylated with acetic anhydride, in a reaction run to leave the phenolic hydroxyl untouched, giving paracetamol directly in a single, high-yielding step.

Diazotisation

The amine group is converted to a diazonium salt under acidic nitrous acid conditions, the first step toward azo dye synthesis via subsequent coupling reactions.

Etherification/Alkylation

The phenolic hydroxyl can be alkylated to give ether derivatives used in specialty intermediate synthesis, while the amine is protected or otherwise accounted for.

Oxidative Coupling

Under controlled oxidation, the amine/hydroxyl pair can couple to form quinone-imine structures, a reaction pathway relevant to both intentional dye chemistry and the unwanted discoloration seen in poorly stored material.

Signal word

Warning

This product is harmful if swallowed and may cause an allergic skin reaction and eye irritation on contact, and is toxic to aquatic life with long-lasting effects, so release to drains and watercourses should be avoided. Routine handling calls for chemical-resistant gloves, eye protection and a dust mask given its fine powder form. Full first-aid, storage, spill and PPE guidance is in the MSDS issued with every shipment, and should be read in full before this material is handled.

07 / STORAGE & SHELF LIFE

Keeping it on spec between delivery and use

General storage guidance for this grade. Site-specific storage rules, segregation requirements and re-test intervals should follow your own EHS procedures and the MSDS issued with your batch.

Storage TemperatureCool, ambient, dark
ContainerDouble-lined drums / fibre drums
Typical Shelf Life18 – 24 months, unopened, dark storage
Keep Away FromLight, air exposure, oxidizers
Beyond Shelf LifeRe-check colour and assay before use

Store in a cool, dark area in tightly closed, light-excluding containers, since exposure to air and light is the principal driver of the slow discoloration this compound is prone to. Keep away from strong oxidising agents. Re-check assay and colour on a batch held past its retest date before use, particularly before committing it to pharmaceutical-grade paracetamol synthesis.
