01 / PRODUCT DOSSIER

What 2,3,4-Trichloro Nitro Benzene is, and why buyers source it

A working overview of the compound's chemistry, its place as a further-chlorinated relative of our dichloronitrobenzene range, and its documented role in quinolone antibacterial synthesis.

2,3,4-Trichloro Nitro Benzene is a benzene ring carrying three chlorine atoms at the 1, 2 and 3 positions and a nitro group at position 4, making it the most heavily chlorinated member of our chloronitrobenzene range. It can be thought of as a further-chlorinated relative of 3,4-dichloronitrobenzene, carrying an additional chlorine at the position adjacent to the existing pair.

At room temperature it is a white to light yellow crystalline powder. It is practically insoluble in water but dissolves in common organic solvents, and like other nitroaromatics is handled as a material requiring full PPE and MSDS-guided handling rather than a benign commodity solid.

Where it fits in the synthesis chain

This compound is documented as a key starting material for third-generation quinolone antibacterials, feeding the same general synthesis family as lomefloxacin and ofloxacin via a related sequence of ring-functionalisation steps to 3,4-dichloro-1,2-benzenediamine-type intermediates. Its activated ortho-chlorines (flanked by both a nitro group and neighbouring chlorines) also make it a documented substrate for halogen-exchange (Halex) fluorination, converting it to 2,3,4-trifluoronitrobenzene — a route represented elsewhere in our range.

Grades we supply

Chelora Chemicals offers 2,3,4-Trichloro Nitro Benzene in technical grade (≈97% assay) for standard industrial synthesis, and high-purity grade (98%+) for pharmaceutical-intermediate and Halex-feedstock applications sensitive to colour or isomeric carryover. Both grades are supplied with a batch-wise certificate of analysis.

02 / MANUFACTURING ROUTE

How 2,3,4-Trichloro Nitro Benzene is made

An overview of the production route for orientation purposes. Our manufacturing partners' own process parameters and in-process controls are proprietary and are not disclosed beyond what is needed to interpret the certificate of analysis.

2,3,4-Trichloro Nitro Benzene is manufactured by further chlorinating 3,4-dichloronitrobenzene, or by nitrating 1,2,3-trichlorobenzene, under controlled Lewis-acid-catalysed or mixed-acid conditions depending on the route selected. Industrially, the choice between these two routes generally comes down to feedstock availability and the isomeric purity achievable at each step.

After the halogenation or nitration step, the crude product is separated from residual acid or catalyst, washed, and crystallised to the required purity and colour before packing.

Why this compound matters as a Halex feedstock

Beyond its role in antibacterial synthesis, this trichloronitrobenzene is the standard starting material for the industrial Halex (halogen-exchange) route to 2,3,4-trifluoronitrobenzene: under halogen-exchange conditions with potassium fluoride, a polar aprotic solvent, phase-transfer catalysis and elevated temperature, all three chlorines are sequentially displaced by fluoride, giving the trifluoro product described elsewhere in our range.

03 / APPLICATIONS

Industries and processes that use it

2,3,4-Trichloro Nitro Benzene is consumed mainly as a pharmaceutical synthesis intermediate and as the standard Halex feedstock for its trifluoro analogue. The fields below account for most of the demand we see through our export desk.

Field 01

Fluoroquinolone Antibacterial Intermediate

A documented starting material feeding the same general synthesis family as lomefloxacin and ofloxacin, via ring-functionalisation to benzenediamine-type intermediates.

Field 02

Halex Fluorination Feedstock

The standard starting material for the industrial halogen-exchange route to 2,3,4-trifluoronitrobenzene, under potassium fluoride / polar aprotic solvent conditions.

Field 03

Fine-Chemical & Specialty Synthesis

Used more broadly as a heavily chlorinated nitroaromatic building block in fine-chemical synthesis where multiple ring chlorines are required.

Field 04

Dye & Pigment Intermediate Chemistry

Its reduction products are used in some dye and pigment intermediate synthesis requiring a trichlorinated aniline building block.

04 / REACTION CHEMISTRY

How the molecule is typically carried forward

The nitro group is the reduction handle, while the three chlorines — particularly those flanked by the nitro group and by each other — are activated toward nucleophilic displacement. Reaction classes below cover the main ways this material is converted downstream.

Halex Fluorination

Under halogen-exchange conditions with potassium fluoride, phase-transfer catalysis and elevated temperature, the three chlorines are sequentially displaced by fluoride to give 2,3,4-trifluoronitrobenzene.

Reduction to the Trichloroaniline

Catalytic hydrogenation or chemical reduction converts the nitro group to a primary amine while leaving the chlorines in place, giving a trichloroaniline intermediate used in fluoroquinolone synthesis.

Further Ring Functionalisation

The reduced amine undergoes further nitration/reduction sequences in the established route toward benzenediamine-type intermediates used in quinolone ring-closure chemistry.

Selective Nucleophilic Aromatic Substitution

Individual chlorines, particularly those most strongly activated by the nitro group's position, can be selectively displaced by other nucleophiles under controlled SNAr conditions.
